For enzyme activity staining of tartrate-resistant acid phosphatase and alkaline phosphatase

Normal bone metabolism is based on the balance between bone formation by osteoblasts and bone resorption by osteoclasts. When the balance is disturbed and bone resorption by osteoclasts is abnormally increased, bone mass is reduced leading to osteoporosis. Therefore, various researches have been carried out to understand the mechanism of osteoclast and osteoblast metabolism, and to utilize this knowledge for the treatment of the diseases and for the development of effective drugs.
Today, alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) are known as marker enzymes for osteoblasts and osteoclasts, respectively, and these enzymes are used as one of the markers showing the presence of osteoblasts and osteoclasts in tissue sections or cultured cells.
This kit enables you to examine the state of differentiation of bone cells and the cell distribution in bone tissues by observation of the stained images of osteoblasts and osteoclasts in the tissues and cultured cells using ALP/TRAP enzyme activities in the tissue sections and cultured cells.

[Features]

In use, just mix two solutions for the preparation of the colorimetric substrate solution required for staining of acid phosphatase enzyme activity. (three solutions in case of the evaluation of tartrate-resistant acid phosphatase)

Simple steps for staining of alkaline phosphatase enzyme activity by using ALP substrate soln. (premixed).

Double staining can be performed, reddish-purple for active region of acid phosphatase and bluish brown for alkaline phosphatase.

Usable in cultured cells and bone tissue sections (GMA resin embedded section).

Hichrom C8 and C18

Hichrom’s C8 and C18 columns offer high performance in order to tackle the most challenging reversed-phase applications. The use of ultra pure silica, advanced bonding technology, superior column specification and comprehensive batch validation all contribute to the columns’ excellent reproducibility.

Excellent Batch Reproducibility

Combining ultra pure silica with advanced bonding technology results in a densely bonded silica that is both highly robust and highly reproducible. Unpredictable interactions between residual silanol sites on the silica surface and the analyte are effectively eliminated. Comprehensive batch validation ensures absolute batch to batch reproducibility is maintained with acidic, basic and neutral molecules. Figures 1 and 2 show two of the tests used during the validation of Hichrom C8 and C18 phases.

Every Hichrom C8 and C18 column is individually manufactured and meticulously tested in the Hichrom laboratory. Efficiency measurements and two peak asymmetry calculations are recorded for each component of the quality control test mixture. Samples of the quality control test mixture are available on request. Only columns exceeding the most stringent efficiency and peak shape specifications are approved for sale.

All Hichrom C8 and C18 columns are supplied with a Test Chromatogram and Batch Validation Certificate. Information regarding column care and use is displayed on the reverse of the Test Chromatogram.

Bonding Stability

Combining a unique dense bonding process (inhibiting ligand cleavage at low pH) with a revolutionary capping process (shielding the silica from dissolution at high pH) enables Hichrom C8 and C18 columns to offer exceptional stability across an extended pH range. Figure 5 demonstrates the acid robustness of a Hichrom 5 C18 column.

Column Robustness

A stable packed silica bed is critical to the long term performance of the column. Hichrom C8 and C18 columns are packed to extremely high efficiencies with excellent peak asymmetries, ensuring that high performance is maintained throughout the column’s lifetime. Figure 6 shows that the performance of both Hichrom 3.5μm C18 and Hichrom 5μm C18 columns remains unaffected after extended use.

C18 column dimensions, to further extend column lifetime by preventing irreversible adsorption and frit blockage at the top of the column. As shown in Figure 7, a fingertight column coupler (HI-081) is used to connect the guard holder (HI-161) to a Hichrom 5 C18 column. Further information on the guard cartridge range is listed on pages 24 and 25.

Advantages

SUMICHIRALTM?OA-SHELL P1 is our first chiral stationary phase using optical active polymer. It is effective for chiral separation of compounds without amino group or carboxyl group,which are generally difficult to separate with conventional chiral stationary phases. Especially, it shows excellent resolution for aromatic chiral alcohol compounds. Core‐shell silica is used as a support and coated with poly (diphenylacetylene) derivative (optically active spiral polymer).

Unlike full porous particles, the core-shell particle has a non-porous core in the center and a thin porous layer outside of the core.

Method development

SUMICHIRALTM?OA-SHELL P1 makes it easy to set the conditions of mobile phase. The scheme of the general method development is shown in the right figure. For neutral compounds such as aromatic alcohols, it is not usually necessary to add special modifiers. For acidic compounds, add trifluoroacetic acid.

  • Do not use the following solvents as they will cause the column to deteriorate : tetrahydrofuran, 1,2-dichloroethane, chloroform, ethyl acetate, acetone, etc.

Standard mobile phase n-hexane/2-propanol(95/5)

Change the concentration of 2 -propanol and adjust the retention time

(concentration of 2-propanol 0~20%)

Change 2-propanol to ethanol

(concentration of ethanol 0~10%)

For acidic compounds etc., add trifluoroacetic acid

Durability test (concentration of trifluoroacetic acid 0.02~0.1%)

SUMICHIRALTM?OA-SHELL P1 has excellent durability despite the use of optically active polymers as chiral selectors. The mobile phase was continuously fed to the column, and the stability was evaluated under the following conditions. As a result, almost no change was observed in separation factor, resolution, retention time, and column pressure.

HICHROM CHIRAL COLUMNS

Hichrom manufactures two ranges of ‘Pirkle-type’ columns for the analysis of a wide range of enantiomers of pharmaceutical or agrochemical origin. CHIRA-chrom 1 type columns contain a stationary phase in which the hydroxyl groups on the silica surface have been modified by a nitrobenzoyl derivative of an optically active amino acid. In the CHIRA-chrom 2 type columns a chiral dinitrophenyltartramide moiety is bonded to the silica surface through a propyl spacer group.

Alltech公司經過多年來生產和革新,成功開發出大規模制造石英毛細管柱技術,對幾類常用氣相色譜固定相的涂漬與固定化,實現了高度的重現性。因此可以在毫不影響質量的情況下,利用批量生產工序,大大降低了生產成本。所有批量生產的經濟型毛細柱,都附有標準的評價報告、譜圖及使用手冊。用戶只需按照使用手冊對其進行老化操作就可正常使用。

Heliflex(R) USP 467 毛細管柱套裝
美國藥典方法467是分析藥物中OVI(有機揮發性溶劑)含量的標準方法。
? 奧泰提供兩種套裝產品,適用于USP 467方法1和方法2,都含有規定的色譜柱和保護柱連接器。

方法1套裝:
AT(TM)-5, (USP G27) 30m x 0.53mm x 5.00μm; 5個連接器; 5 x 5m保護柱 (0.53mm ID)。

方法2套裝:
AT(TM)-624, (USP G43) 30m x 0.53mm x 3.00μm; 5 連接器;5 x 5m 保護柱 (0.53mm ID)。

AT(TM)-Petro 毛細管柱有著相對低的固定相比率,分析輕石油產品有著很高的柱效(分析異丁烷/正丁烷以上產品),平均理論踏板數為400,000 /每根,高容量性允許極濃的樣品直接進樣,是典型的石油樣品分析柱。

Heliflex(R)?? Chirasil-Val(R)

? 交合鏈接,保證固定相的熱穩定性
奧泰Chirasil-Val(R)是采用專業的分離光學異構體的固定相,這種手性固定相已經成為分離測定氨基酸尤其是合成縮氨酸工業中監控外消旋異構體含量的標準固定相。這種固定相有很多重要的應用,比如測定考古發現中測定外消旋氨基酸方法、分析糖異構體和手性藥物的分析(包括R,S-Mephenytoin和R,S-Tocainide)。

Chirasil-Val(R)? 是采用手性功能團鍵合在硅樹脂上,zui高使用溫度為等溫220℃ ,梯度升溫240℃ 。我們保證每根色譜柱出廠前都經過混合氨基酸的測定,都具有高穩定性和高柱效。 Chirasil-Val(R)沒有通用的固定相, 只是毛細管涂層。

Alltech PLOT 毛細管柱
現在許多氣相色譜分析方法逐漸用高柱效毛細管柱替代了低柱效的氣相填充柱,從而得到更好的分離度和更好的重現性,但是在某些領域比如揮發性氣體分析,常用氣體分析,低沸點溶劑等等。在這些分析中只有選擇固定相涂層膜非常厚的毛細管柱,但是仍有很多氣體仍不能得到良好的分離,只有采用填充有多空高分子小球、分子篩和三氧化二鋁填料的填充柱?,F在奧泰公司能夠將這些填料填充到0.53mm 內徑的石英毛細管柱內,PLOT (多孔層開關柱)毛細管柱可以用于毛細管分析儀器或經過簡單改裝填充柱分析儀器 (需改裝儀器請與我公司技術工程師), 并可用于您分析系統的任何一種檢測器 (FID, TCD, MSD, 等等)。
